Dit is het tweede deel van de Professor Layton trilogie voor de Nintendo DS waarin de gelijknamige professor de hoofdrol speelt. De game is een spel waarin je vooral puzzels moet oplossen. De game bezit meer dan tweeduizend puzzels die het touchscreen en de stylus gebruiken.
