Met Resistance 3 gaat het verhaal over de invasie van de Chimera verder. Amerika is volledig ingenomen door de mutanten en jij als Joseph Capelli bent als één van de weinige mensen overgebleven.
De game brengt je een nieuwe Trophy-lijst, bestaande uit 59 Trophies. De meerderheid daarvan, 53 om exact te zijn, zijn bronzen trophies. Er zijn eveneens vier zilveren, één gouden en één platinum trophy te verdienen, samen goed voor 1245 punten. Deze trophies concentreren zich op singleplayer en coöp, competitieve multiplayer-Trophies zijn er ditmaal niet te verdienen.
